The Role of IL-18, CCL5 and their receptors as Diagnostic Markers in Breast Cancer

Interleukin-18 (IL-18) is a proinflammatory cytokine that has been suggested to be a prognostic factor in some cancer types.  Also, CCL5 has been considered as a potential contributor to breast cancer progression. Using RT-PCR, this workexamined the levels of IL-18, CCL5 and their receptors in patients with breast cancer (BCa) in Bahrain. In this study we compared tissueswith and without metastasis to tissues from benign and normal breast. Formalin-fixed paraffin-embedded breast tissues wereobtained from 25 patients without metastasis, 20 patients with metastasis, 15 with benign tumor and 15 with normal breast tissues (total=75). IL-18/IL-18R expression in metastatic and non-metastatic group was significantly high when compared to control group (p<0.05).  IL-18/IL-18R expression in the metastatic BCa was higher than the non-metastatic BCa, but did not reach statistical significance.  The expression of CCL5/CCL5R in metastatic and non-metastatic group was significantly high when compared to control group (p<0.05). As IL-18/IL-18R, the expression of CCL5/CCL5R in the metastatic patients was higher than the non-metastatic, but as well did not reach statistical significance. In conclusion, IL-18/IL-18R and CCL5/CCL5R may be useful markers in the detection of malignancy in breast, however, their role in the spread of the disease and metastasis needs to be further investigated.
